Double-stranded nature of RCNMV replicase products
RCNMV replicase products are double-stranded RNA
- RNase A digestion of RCNMV replicase products. Fractions of replicase were assayed in the
absence of exogenous templates. RCNMV ss genomic RNA (lane 3), and replicase products of S100
fraction (lane 1) and F3 fraction (lane 2) were fractionated on a 1.2% native agarose gel and subjected
to 50 mg/ml RNase A digestion for 1 hr. in 2X SSC (B), 0.2X SSC (C), and no RNAse A in 0.2X SSC (A).
Bottom panel: ethidium bromide stained picture after the RNase A digestion showing RCNMV RNA
was digested in both 2X SSC and 0.2X SSC. Top panel: The same gel slices were dried and
autoradiographed for 15 hrs at -70 C. Notice that the replicase products were digested by RNAse A in
0.2X SSC, but not in 2X SSC.
